AAgbas
Hello mito community, I am curious about the aging mark on mitochondria. How can I asses that mitochondria is aging other than enzymatic activities of mito-enzymes? Thanks for your thoughts
pdacks
You could quantify mitochondrial DNA damage, perhaps at particular sites of the DNA? Are you looking in tissue or cell culture?
Jaime_Ross
Have you been successful in determining a method? To my knowledge, “ageing” mitochondria are typically associated with mitochondrial dysfunction, so perhaps that is why the typical approach is to measure the activities and amounts of Complexes I-V. Perhaps investigating mtDNA copy number or the level of somatic mtDNA mutations would also be helpful.
AAgbas
Many thanks. I do have some of enzymatic assesement protocols, however, I am in need of Complex I-II combination assay method. Can anybody provide me one? Regards
